Diagnostic Biosystems BCL-2 ONCOPROTEIN Bcl-2 Oncoprotein
This antibody recognizes a protein of 25-26kDa, identified as bcl-2 alpha oncoprotein. It shows no cross-reaction with Bcl-x or Bax protein. Expression of bcl-2 alpha oncoprotein inhibits the programmed cell death (apoptosis). In most follicular lymphomas, neoplastic germinal centers express high levels of bcl-2 alpha protein, whereas the normal or hyperplastic germinal centers are negative. This antibody is valuable when distinguishing between reactive and neoplastic follicular proliferation in lymph node biopsies. It may also be used in distinguishing between those follicular lymphomas that express bcl-2 protein and small number in which neoplastic cells are bcl-2 negative.

Pepsin is used for proteolytic digestion of formalin-fixed paraffin-embedded (FFPE) tissue sections prior to application of antibodies. In immunohistochemistry (IHC), most commonly used fixatives such as formalin mask tissue antigens (cellular, membrane, and nuclear) by their intrinsic crosslinking. This masking results in poor or no staining in IHC. Pepsin digestion of FFPE tissue sections improves accessibility of antibodies to tissue antigens.\n\nFixation is one of the most critical aspects of immunostaining. If the antigen is not properly fixed it will be washed out of the specimen. On the other hand, overfixation can cause severe problems such as masking or denaturation of the antigen. Formaldehyde is probably the best all-around fixative, due to its cross-linking characteristics. Length of fixation is very critical to prevent antigen masking.

This product is used as a signal enhancer in conjunction with alkaline phosphatase based immunostaining systems.\n\nAlkaline Phosphatase, an enzyme derived from bovine intestinal mucosa, is often used as a label for in situ hybridization, immunohistochemistry, Southern and Northern hybridization and DNA sequencing. Detection of this enzyme requires the generation of an insoluble colored reaction end product. Stronger the signal, better the staining. DBS introduces a new stable solution to increase the signal generated by alkaline phosphatase several fold. A single treatment of tissue sections with enhancer before the treatment with substrate/chromogen enables the visualization of difficult to localize antigens. This enhancer works well with Fast Red but can be used with BCIP/NBT and BCIP/INT also. Specimens stained with the above substrates/chromogens can not be dehydrated in ethanol and hence should be mounted in aqueous based mounting medium like C/C Mount (DBS catalog # K002).
